Tocopherols Inhibits the Onset of Colon Cancer in Vitro

The colon also is known as the large bowel or large intestine, formed parts of the digestive system in our body.

The colon consists of four parts: descending colon, ascending colon, transverse colon, and sigmoid colon.

The main functions of the colon are to reabsorb fluids and process waste products from the body and prepare for its elimination.

Colon cancer is a chronic condition caused by cell growth irregularly and disorderly in the colon due to the alternation of DNA.

Most cases of colon cancer start in the cells on the surface of the inner lining of the colon. The stage of colon cancer is depended on how far the alternated cells have spread in the colon.

Patients with the early stage of colon cancer, may not experience any symptoms. But as the tumor spread and expanses, it may suppress the nearby tissue and nerve, causing symptoms of locally severe pain and discomforts.

Although there are many risk factors associated with the early onset of colon cancer, a poor diet high in saturated fat and trans fat, red meat and processed foods have been found to play a major role in the development and recurrence of colon cancer in the Western world.

According to the statistic by the American Cancer Society, in the US, there are
97,220 new cases of colon cancer were diagnosed in 2018. The lifetime risk of developing colorectal cancer is 1 in 22 (4.49%) for men and 1 in 24 (4.15%) for women.

The causes of colon cancer are unknown but certain risk factors, such as genetic preposition and chronic inflammation induced by colonic diseases are associated with the onset of the disease.

According to the statistics provided by the Canadian Cancer Society, In Canada, the 5-year net survival for colorectal cancer is 65%.

In other words, out of 100 people diagnosed with colon cancer, 65% of people will survive for at least 5 years after their diagnosis.

Tocopherols are phytochemicals of which many have vitamin E activity, belonging to the group of Lipids, found abundantly in butter, egg yolk, milk fat, some vegetable, and seed or nut oils, etc.

On finding a potential phytochemical for the treatment of chronic diseases, researchers examined the effect of co-treatment of γ-tocopherol and 5-FU in enhancing the efficacy of chemotherapy against HT-29 colon cancer cells.

According to the analysis,
* The IC50 values for 5-FU and γ-tocopherol were 21.8  ± 2.5 and 14.4 ± 2.6 μM, respectively, and also this combination therapeutic increased the percentage of apoptotic cells from 35% ± 2% to 40% ±  4% (P  <  .05).
* Incubation HT-29 colon cells with combined concentrations of two drugs also caused significant accumulation of cells in the subGsubG1 phase.

After talking about other factors into account, researchers said, "Our results presented the combination therapy with 5-FU and γ-tocopherol as a novel therapeutic approach, which can enhance the efficacy of chemotherapy".

Taken altogether, tocopherols may be considered an adjunct therapy in the treatment of colon cancer, pending the confirmation of the larger sample size and multicenter human study.

Intake of tocopherols in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Beta-Sitosterol, a Natural Free Radical Scavenging Antioxidant

Antioxidants are stable atom that plays a critical role in protecting the body against the attack of free radicals.
In other words, antioxidants inhibited the onset of free radicals before they can cause a chain reaction in facilitating protein, lipid, and cell damage.

Antioxidants can either be produced by the liver or intake from dietary sources. Under normal conditions, antioxidant enzymes produced by the body in a healthy individual are good enough to counter the bombardment of free radical attacks.

However, in people with a weakened immune system, intake of dietary sources of antioxidants may be necessary.

Free radicals are highly reactive molecules with an unpaired electron on the outer ring of the atom.
Due to its nature to become stable, free radicals readily donate or steal an electron from other molecules, leading to the chain of domino's effect that can not be stopped until either the electrons are paired or inhibited by antioxidants.

This chain of reaction is the cause of oxidative stress, a condition is associated with either overexpression of free radicals or depletion of antioxidant enzymes produced by the host organs.

Overexpression of free radicals is known to cause damage in contributing to the etiology of many chronic health problems such as cardiovascular and inflammatory disease, cataracts, and cancer.

The expression of oxidative stress is a medical condition characterized by the imbalance of the ratio of free radicals and antioxidant enzymes produced by the host tissue.

In some cases, this reaction also is caused long-term accumulation of toxic substances in the body, such as lead, cadmium, mercury, and arsenic.

Beta-Sitosterol is a phytochemical in the class of Phytosterols, belongings to the group of Lipids, found abundantly in avocados, rice bran, wheat germ, corn oils, fennel, peanuts, soybeans, hawthorn, basil, buckwheat. etc.

On finding a potential phytochemical for the prevention and treatment of diseases associated with oxidative stress, researchers elucidated in vitro antioxidant and antimicrobial potential of Schimpera arabica extract and fractions (2-Hexadecen-1-ol and beta-sitosterol).

According to the tested assays,* Among all the extracts, 2-Hexadecen-1-ol and beta-sitosterol were the major constituents of crude extract and exhibited significant antioxidant potential as well antimicrobial activity, compared to all other fractions that only exhibited a fair degree of antioxidant potential.
* In the free radical assay, 93% DPPH was scavenged by 100 μg ml-1 of the ethyl acetate fraction; i

* In ABTS assay chelation was 86·2 and 88% in ferric chloride reducing power assay.

Based on the findings, researchers said, "S. arabica contains a good amount of natural antioxidants of medicinal importance. However, further investigations are needed to characterize the further activity of 2-Hexadecen-1-ol and beta-sitosterol in vivo.".

Taken altogether, beta-Sitosterol found abundantly in S. arabica may be considered an antioxidant supplement, pending the confirmation of the larger sample size and multicenter human study.

Intake of beta-Sitosterol in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Beta-Sitosterol Improves The Liver and Kidney Functioning in Hypertensive Rats in Vivo

Blood pressure is the force of the blood pushing against the arterial walls. In high blood pressure, the heart has to work harder than normal to pump blood through the circulation system. Over time, high blood pressure has been found to cause damage to the heart and arteries.

Hypertension or high blood pressure is a condition characterized by persistently or abnormally high blood pressure that leads to raising the pressure in your heart.

It also is a member of the cluster of metabolic syndrome associated with the risk of diabetes and cardiovascular disease.

High blood pressure is a slowly developed condition. Unless the syndrome has caused damage to the blood vessel, otherwise symptoms cannot be detected.

If the pressure stays high over time it can cause hardening and stiffening arterial wall,, a major risk factor of heart diseases.

The ideal blood pressure is more than 120 over 80 and less than 140 over 90 (120/80-140/90), according to the consensus.

In the US approximately, one-third of the adults have high blood pressure, affecting over 77 million people in the country.

Additionally, among adults age 20 and older in the United States, in the non-Hispanic whites community, 33.4 percent of men and 30.7 percent of women are infected by the syndrome.

High blood pressure can be acute or chronic. Acute high blood pressure most often occurs in people with sudden stress or intense physical exercise. As the heart must pump harder to provide the needs of nutrients and oxygen.

Most cases of chronic high blood pressure are associated with an unhealthy lifestyle and chronic diseases such as kidney disease, leading to the onset of strokes and cardiovascular disease.

If you can control your diet, such as making changes to your lifestyle, maintaining an ideal weight, and diet with low salt, you could do just fine without the drugs.

Beta-Sitosterol is a phytochemical in the class of Phytosterols, belongings to the group of Lipids, found abundantly in avocados, rice bran, wheat germ, corn oils, fennel, peanuts, soybeans, hawthorn, basil, buckwheat. etc.

On finding a potential phytochemical for the prevention and treatment of metabolic syndrome, researchers compared the effects of β-sitosterol (BSS) and lisinopril, antihypertension in rats.

According to the tested analysis of rat groups induced hypertension by cadmium chloride treated by BSS and lisinopril, * BSS showed a significant decrease (p<0.05) in the serum creatinine concentration, compared to hypertensive rats.

* Lisinopril treatment showed a significant increase (p<0.05) in the activity of oxidative stress markers AST and ALP, compared to normal rats.

* Treatment with BSS and lisinopril showed slight variations in the concentration of serum electrolytes compared with normal and hypertensive rats.

* BSS significantly reduced calcium levels compared with the hypertensive group in the activation of oxidative stress.

Based on the findings, researchers said, "The study shows that BSS is effective in restoring basal liver and kidney functions in hypertensive rats.

Taken altogether, beta-Sitosterol may be considered a supplement for the protection of liver and kidney functioning, pending the confirmation of the larger sample size and multicenter human study.

Intake of beta-Sitosterol in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Omega-3 Fatty Acids Promote Neuro-Functioning After Ischemic Stroke

Cardiovascular diseases refer to heart diseases and stroke, affecting millions of people in the world.
According to statistics, over 17.7 million people died from CVDs in 2015, Hyperlipidaemia and hypertension are the risk factors of the diseases.

In other words, stroke can be caused by either ischemia, due to lack of blood flow or hemorrhagic, due to bleeding.

A stroke is a medical emergency. Early intervention is most cases can minimize brain damage and potential complications.

Not all strokes can lead to permanent brain damage. Some people with a temporary disruption of blood flow to the brain do not result in permanent damage.

Besides aging, smoking, medical conditions such as unmanaged high blood pressure and cholesterol and diabetes, obesity, physical inactivity are some of the prevalent factors found in stroke patients.

There is a general belief that stroke is an accumulation over a long period of an unhealthy diet with red meat and processed meats in the induction of artery-clogging.

Some researchers suggested that increased intake of fruits and vegetables such as carrot accompanied with good fats such as salmon, tuna, and mackerel may have a substantial effect in reduced inflammation in the arteries, improved blood flow, and decreased the chance of blood clots.

Sadly, stroke is one of the most common causes of death worldwide.

In the US, according to statistics, approximately 800,000 people suffer a stroke of which 600,000 of these are first attacks, and 185,000 are recurrent attacks. Nearly three-quarters of all strokes occur in people over the age of 65.

According to the statistics, in the US, stroke is the leading cause of serious, long-term disability.

Omega-3 fatty acids are phytochemicals in the class of lipids, found abundantly in deepsea fisk, dark-green leafy vegetables, grains, legumes, nuts, etc.

The three main components of Omega-3 fatty acids are alpha-linolenic acid (ALA), eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA).

On finding a potential phytochemical for the treatment of cardiovascular diseases, researchers examined the effects of mega-3 polyunsaturated fatty acids (n-3 PUFAs) in alleviating ischemic brain injury in young adult rodents.

The study included aged (18-month-old) mice which were administered n-3 PUFA-enriched fish oil in daily chow for 3 months before and up to 8 weeks after 45 minutes of transient middle cerebral artery occlusion (tMCAO).
According to the results of differentiation,
* Mice receiving dietary supplementation of n-3 PUFAs for 3 months showed significant increases in brain ratio of n-3/n-6 PUFA contents.

* Furthermore, the administration of dietary supplementation of n-3 PUFAs for 3 months also markedly reduced long-term sensorimotor deficits and chronic ischemic brain tissue loss after tMCAO.

* Moreover, n-3 PUFAs robustly promoted post-ischemic angiogenesis and neurogenesis to enhance the brian neuron activity and enhanced white matter integrity after tMCAO.

* Most importantly, the enhancement of neurogenesis and white matter integrity were both correlated positively with the improvement of sensorimotor activities after tMCAO.

Collectively, researchers said, "prophylactic dietary supplementation of n-3 PUFAs effectively improves long-term stroke outcomes in aged mice, perhaps by promoting post-stroke brain repair processes such as angiogenesis, neurogenesis, and white matter restoration".

Taken altogether, Omega-3 fatty acids found abundantly in fish oil and plant oil may be considered a functional food for the improvement of long-term stroke outcomes, pending the confirmation of the larger sample size and multicenter human study.

Intake of Omega-3 fatty acids in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.
