Antioxidant Omega 3 Fatty Acids Process Cardio-Protective Activity in Vivo

Cardiovascular diseases (CVD) are a class of conditions associated with the dysfunction of the arteries and heart, affecting heart functioning.

According to the statistics provided by the CDC, heart disease is the leading cause of death for both men and women. Men are associated with 1/2 of the deaths due to heart disease in 2015 were in men, compared to women.

Believe it or not, more than 63),000 Americans die from heart disease each year or 1 in every 4 deaths are caused by heart disease.

On another hand, stroke is a medical condition caused by poor blood flow to the brain, leading to brain cell death in the affected region.

Cardioprotection is an action that contributes to the protection of heart and blood vessels integrity against damage or injury, including heart diseases, and stroke.

In other words, cardioprotection is a process in which substances or medicines are used including an herbal remedy to protect heart muscle tissues against ischemic insult and deoxygenation injury.

Coronary artery disease caused by narrowing of the arteries, heart attack, abnormal heart rhythms, or arrhythmias and stroke is the most common type of cardiovascular disease found in the Western world.

The most common symptoms of CVD include persistent fatigue, shortness of breath, irregular heartbeat, chest pain, and fainting.

If you have some of the aforementioned symptoms, please make sure that you check with your doctor to rule out the CVD possibility.

The exact causes of cardiovascular diseases are unidentified. However, according to epidemiological studies, the most prevalent factors include aging, smoking. lack of exercise. unhealthy diet. obesity. high blood pressure and cholesterol, and family history.

Omega-3 fatty acids are phytochemicals in the class of lipids, found abundantly in deepsea fisk, dark-green leafy vegetables, grains, legumes, nuts, etc.

The three main components of Omega-3 fatty acids are alpha-linolenic acid (ALA), eicosapentaenoic acid (EPA), and docosahexaenoic acid (DHA).

On finding a potential phytochemical for the treatment of cardio-diseases, researchers examined the effects of 3 diets with different omega-3/6 fatty acid ratios on infarct size on the lipid composition of cardiac tissue.

According to the results of Sprague-Dawley rats that were fed omega-3/6 fatty acid diets with 1:1, 1:5, or 1:20 ratios for at least 10 days, followed by occlusion of the left anterior descending artery for 40 min and 24 h of reperfusion,* The infarct size was significantly smaller in the 1:1 group than in the other groups.
* The significantly higher concentrations of the omega-3 fatty acids eicosapentaenoic acid, docosapentaenoic acid, and docosahexaenoic acid were found in the 1:1 group than in the other groups.

* Omega-6 polyunsaturated fatty acid levels were similar between groups, although they were higher in the 1:5 and 1:20 groups than in the 1:1 group.

* Docosahexaenoic acid levels in cardiac tissue and infarct size were significantly correlated with no other significant links being apparent.

Moreover, researchers said, " The present study indicated that a 1:1 omega-3/6 fatty acid ratio protected against ischemia and was associated with the increased omega-3 fatty acid composition of cardiac tissue".

Taken altogether, Omega-3 fatty acids may be considered a cardio-protective remedy, pending the confirmation of the larger sample size and multicenter human study.

Intake of Omega-3 fatty acids in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Phytochemical α-Tocopheryl succinate (α-TOS) Protects the Brain Against the Onset of Cerebral Malaria in Vivo

The brain-formed part of the central nervous system is the most complex organ made up of more than billions of nerve cells or neurons.

The functions of the brain are associated with the communication of trillions of connections called synapses in many specialized regions.

A neuron or nerve cell made of a cell body (also called soma), dendrites, and an axon, is a cell in the nervous system that carries electrical impulses.

Neurons are core components of the brain, spinal cord, and peripheral
Neurological disorders are diseases of the central nervous system including the brain, spine, and the nerves that connect them. The most common diseases associated with neurological disorders in the elderly are dementia.

Cerebral malaria is the most severe case of neurological complications caused by the infection with the Plasmodium falciparum malaria and parasite on peripheral blood smears.

The most common symptoms of cerebral malaria are fever, headache, body ache, and progressively, delirium and coma.

However, in adults, cerebral malaria is part of a multi-organ disease.

In African children, seizures papilledema, and retinal changes are less common and coma resolution is slower, compared to other races.

Conventionally, the first-line treatment of cerebral malaria is therapy with concentrations of anti-malarial drugs as quickly as possible.

Tocopherols are phytochemicals of which many have vitamin E activity, belongings to the group of Lipids, found abundantly in butter, egg yolk, milk fat, some vegetable, and seed or nut oils, etc.

On finding a potential phytochemical for the treatment of cerebral diseases, researchers examined the effects of α-Tocopheryl succinate (α-TOS), a derivative of vitamin E, synthesized by esterification of α-tocopherol for treatment of cerebral malaria.

On C57BL/6J mice infected with P. yoelii 17XL and P. berghei ANKA, a lethal strain of rodent malaria, treatment with several concentrations of α-TOS by intraperitoneal administration on 1, 3, 5, and 7 days post-infection (dpi) showed a strong effect on Plasmodium parasites by oxidative stress.

Moreover, α-TOS treatment decreased the parasitemia and increased the survival rate significantly in mice infected with both parasites.

Additionally, the intensity of Evans blue staining on brains taken from α-TOS-treated mice was weaker than that of untreated mice.

In other words, α-TOS inhibited the breakdown of BBB that allow the initiation and progression of cerebral malaria.

Collectively, researchers said, "These findings indicate that vitamin E derivatives like α-TOS might be a potential candidate for treatment drugs against malaria".

Taken altogether, tocopherols may be considered a supplement for the prevention and treatment of cerebral malaria, pending the confirmation of the larger sample size and multicenter human study.

Intake of tocopherols in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.

Omega 3 Fatty Acid Promotes Myocardial Ischemia-Reperfusion in Vivo

Compared to acute brain infarction that is a medical condition characterized by the obstruction of the blood supply to the brain, causing local death of the brain tissue. The occurrence and extent of brain infarction are associated with three basic factors, including the site of arterial occlusion, the rapidity of arterial occlusion, and the presence or absence of collateral circulation.

Myocardial ischemia injury is a medical condition caused by severe impairment of coronary blood supply induced damage or dysfunction of the cardiac tissue.

Myocardial reperfusion is the process to restore the coronary blood flow after a period of coronary blockage that leads to severely reduced or no oxygen .blood flow to the heart.

The condition severity is totally dependent on the intensity and duration of the coronary occlusion.

According to the statistics from the WHO, in 2008, 12% of all death or approximately 7,254,000 deaths worldwide have directly or indirectly caused by myocardial ischemia injury.

Atherosclerosis, one of the major causes of cardiovascular disease is a medical condition caused by the accumulation of plaque on the arterial wall. Over time, the condition can lead to narrowing the arteries or blocked arteries, leading to reduce oxygenated blood flow to the heart and other parts of the body.

Arteries are blood vessels carried the oxygen-rich blood to the heart before passing them to the tissues and organs.

A plague is made up of fat, cholesterol, calcium, and other substances found in the blood. In other words, cholesterol that cannot be returned to the liver for reuse may have a strong impact on inducing plaque accumulation.

Coronary heart disease causes 1/4 total death in the worldwide population and 50% of the death associated with other forms of heart disease.

On finding a potential phytochemical for the treatment of post-stroke, researchers examined the cardioprotective effects of ω-3 polyunsaturated fatty acids (PUFAs) postconditioning against ischemia-reperfusion (I/R) injury.

The study included 60 Sprague-Dawley rats that were randomly divided into 4 groups (n = 15 for each) and used to generate the Langendorff isolated perfused rat heart model.

* The sham group received continuous perfusion of 150 min.

* The remaining three I/R-treated groups sequentially received 30-min perfusion, a 30-min cardioplegia, and 90-min reperfusion.

* The I/R-ischemic preconditioning (IP) group additionally received three cycles of 20-s reperfusion and 20-s coronary re-occlusion preceded the 90 min of reperfusion.
* The I/R-ω group was perfused with ω-3 PUFAs for 15 min before the 90 min of reperfusion.

According to the results of the study,
* Compared with the I/R group, the I/R-ω group had significantly reduced myocardial infarct size, reduced levels of lactate dehydrogenase and malondialdehyde associated with cardio-stress markers.

* Furthermore, Omega 3 fatty acids elevated superoxide dismutase level involved in antioxidant production and elevated rising (+dp/dtmax) and descending (-dp/dtmax) rate of left ventricular pressure against myocardial ischemia-injury.

* Moreover, The I/R-ω group had a significantly lower rate of mitochondrial damage in myocardial tissue compared with the I/R and I/R-IP groups.

In other words, Omega-3 fatty acids protect the heart tissues and muscles against damage after stroke.

Moreover, researchers said, " ω-3 PUFA postconditioning possesses good cardioprotective effects and may be developed into a therapeutic strategy for myocardial I/R injury".

Taken altogether, Omega-3 fatty acids may be considered a remedy for the treatment of myocardial ischemia injury, pending the confirmation of the larger sample size and multicenter human study.

Intake of Omega-3 fatty acids in the form of supplements should be taken with extreme care to prevent overdose acute liver toxicity.
