In animal study, nicotinamide in doses of 50, 100 and 200 mg/kg applied to gestational diabetic fed to rats from days 6 through 20 of gestation showed a significant in reduced diabetes symptoms through increased production of antioxidants, reduced inflammatory cytokins and genes in controlling expression of free radicals.

The doses of 100 and 200 mg/kg t showed an extensive effect against the oxidative burst activity of neutrophils in response to cell differentiation of phorbol myristate acetate (PMA) and N-formyl-methionyl-leucyl-phenylalanine (FMLP) in elevated risk of diabestes complications as well as activating T helper (CD4+CD25+) cells and inducing proliferation of splenocytes, a type of white blood cell involved autoimmune diabetes mellitus expression.

The findings propsoed that nicotinamide as supplement may be used as therapeutic agent in support  existing treatment for gestational diabetes and pregnant individuals with weakened immune systems.

Other in a randomized, double-blind clinical trial, patients with moderate inflammatory facial acne vulgaris randomly allocated to receive either topical 4% nicotinamide (n = 40) or 1% clindamycin gels (n = 40) twice daily, group treated with topical 4% nicotinamide showed an  acne grade decreased from an average of 5.93 at baseline to 4.03 at week 4 and 2.08 at week 8 in compared to
5.70 at baseline to 3.85 at week 4 and 2.03 at week 8 in the clindamycin group.

Further consultation insisted that skin types of patients may play an important role in choosing products as nicotinamide and clindamycin gels were significantly more efficacious in oily and non-oily skin types, respectively.

Both products showed insignificantly adverse side effects during the course of study.

Due to the limited nature of the available literature, some researchers suggested that additional studies are needed in comparing nicotinamide to other first-line acne treatments over an extended period of time may be necessary.

All About Vitamins: Niacin in reduced Risk Oxidative Stress

Niacin, is also known as vitamin B3, nicotinic acid, an organic compound with the formula
C6H5NO2, found abundantly in chicken, beef, fish, cereal, peanuts and legumes. It is best known for its effects in lowering cholesterol and triglycerides and removing toxic from our body and promoting production of steroid hormones.

Niacin may be considered as dietary supplement in reduced risk of oxidative stress in induction of various diseases, a respectable institute study opinionated.

In a total of 127 men and women aged 48-77 yr with brachial artery flow-mediated dilation (FMD), patients, above-average dietary niacin intake showed a significant greater function in compared to those below-average intake (P < 0.05).

The study also found that above-average dietary niacin intake patients showed a lower levels of oxidative stress through expression of plasma oxidized low-density lipoprotein.

Further more, levels of antioxidants also has linearly association to amount intake of niacin as shown in this participants.

Dr. Kaplon RE, the lead author said, " higher dietary niacin intake is associated with greater vascular endothelial function related to lower systemic and vascular oxidative stress among healthy middle-aged and older adults."

In support of the above analysis, the study of 17 patients with hypercholesterolemia and low HDL-C and 8 healthy control subjects, 12 weeks.of niacin treatment showed a strong improvement of lipid profile, oxidative stress and C-reactive protein (CRP) levels, through increase in HDL-C and apolipoprotein A1 levels involving transferred to HDL in the bloodstream and a decrease in triglyceride levels.

More promisingly, Niacin treatment in these patients with low HDL levels showed a significant in reduced oxidative stress through improved activities in regulated C-reactive protein (CRP) levels and lipid profiles.

Taking altogether, There is no doubt that niacin may be considered as a dietary treatment in reduced risk of diseases caused by oxidative stress.

Niacin supplement may have a potential effect in reduced risk and treatment glaucoma, a Pennsylvania study suggested.

Intake of 500 mg of oral niacin(also known as vitamin B3 or nicotinic acid) daily is shown to increased intraocular pressure to 37 and 27 mm Hg, according to the the Jefferson Medical College. study.

In the exam of 73-year-old man with a history of primary open-angle glaucoma, intake of niacin have a significant effect on the eye pressure which may be increased or decreased depending to amount of niacin intake.

The increased abnormal eye pressure also reduced to normal once niacin intake was stopped.

The finding is contrasted to the investigation of mice treated with nicotinamide adenine dinucleotide (NAD), in genetically predisposed to developing glaucoma with healthy mice acted as control, in indication of niacin effect in treatment of glaucoma through anti oxidative stress by Dr. John and colleagues. 

According to Dr. John and colleagues study, the depletion of NAD weakens the metabolism of the brain cells, and intraocular pressure may have a direct influence on the onset of glaucoma. And  in glaucoma-predisposed mice, adding niacin to the drinking water showed to improve aging retinal ganglion cells and modify the neuro change associated with advancing age and prevent the early onset of glaucoma.

Taking altogether, intake of niacin supplement to reduced risk and treatment of glaucoma may need further studies by larger human trial and involvement of multi centers before confirming.

Kidney disease is a medical condition of kidney impairment, characterized by adequately filter metabolic wastes from the blood.

Niacin may have a potential effect as phosphorus lowering agents in patient with ischemic nephropathy, inducingvarious types of syndromes and diseases,  a joint study suggested.

According to the University of Karachi, Karachi, niacin may be one of low cost supplement used management effectively in attenuated elevated serum phosphorus in patient population with compromised kidney performance. Thus reducing co-morbidities especially cardiovascular disease risk and derangement in calcium-phosphorus homeostasis in advancing to onset of chronic kidney disease.

In the study of 170 chronic kidney disease patients and 50 healthy control, fulfilling the pre-defined criteria, randomly categorized into two groups of equal size (n=60) and prescribed either atorvastatin 20 mg/day or extended release niacin(NER) 500 mg/day with the same dose of statin for four months, injection of atorvastatin alone and NER showed a significantly reduced phosphorus level.

In compared to the adverse effect in both treatments, the university also said, "NER, at its lowest prescribed dose once a day was well tolerated by most of the patients and demonstrated significant goal achievement of phosphorus reduction".

Furthermore, in data analysis of serum phosphorus concentrations, among 1547 patients with dyslipidemia and randomly assigned in a 3:2:1 ratio to treatment with extended release niacin (ERN; 1 g/d for 4 weeks and dose advanced to 2 g/d for 20 weeks) combined with the selective prostaglandin D2 receptor subtype 1 inhibitor laropiprant (L; n = 761), ERN alone (n = 518), or placebo (n = 268), repeated measurement of ERN-L treatment resulted in lowering serum phosphorus change comparing ERN-L with placebo.


Dr. Maccubbin D, the lead author siaid, "once-daily ERN-L treatment causes a sustained 0.13-mmol/L (0.4-mg/dl) reduction in serum phosphorus concentrations, approximately 10% from baseline".

Taking together, niacin may be effective for enhanced kidney function by reduced level of phosphorus in both healthy and chronic kidney disease patients. intake of large amount should be taken with care under close observation by medical professional.

According to the Dalian University, co-administration of glucose and 300 mg nicotinamide in animal study showed a significant elevated levels of RSO and insulin levels but the 3-h blood glucose test showed the levels of was significantly lower.

The study result indicated that niacin not only has an appetite-stimulating effect involved oxidative stress but also be concerned as a major factor in the increased obesity prevalence in US children with excess consumption.

Dr.Li D, the lead author said, "The obesity prevalence among American children increased with the increasing per capita niacin consumption, the increasing grain contribution to niacin due to niacin-fortification, and the increasing niacin-fortified ready-to-eat cereal consumption, with a 10-year lag".

Furthermore, in other same subject study conducted by the same university, intake of excess amount of vitamin supplement and fortify food have found to accompany by an increased prevalence of obesity, a condition associated with diabetes, hypertension, cardiovascular disease, asthma and cancer.

Recent experiment also suggested that not only excess niacin showed to induced obesity, obese patients also exhibited low fasting serum but high erythrocyte folate concentrations, exhibiting folate oxidation involved oxidative stress. 

Dr. Zhou SS, the lead author said, "here is also evidence that obesityincreases niacin degradation, manifested by increased activity/expression of niacin-degrading enzymes and high levels of niacin metabolites" and " that low fasting serum vitamin status in obesity may be a compensatory response to chronic excess vitamin intake, rather than vitamin deficiency, and that obesity could be one of the manifestations of chronic vitamin poisoning".

The finding evidenced proposed that excess vitamins intake may have a potential toxic effect in obese patients.

In the review of 72,308 women in the Nurses' Health Study (1984-2010) and 41,808 men in the Health Professionals Follow-up Study (1986-2010), with niacin intake assessed every 2 to 4 years during follow-up and cumulative averaged intake,  niacin injection was inversely associated with risk of skin cancer in total documented 23,256 BCC, 2,530 SCC and 887 melanoma cases.

The investigation also addressed the gender different in higher total niacin intake and risk of melanoma.  As men is found to marginally positively associated with melanoma risk, but not in women.

Also, according to the study, niacin may be effective in reduced risk and treatment of skin cancer of type squamous cell carcinoma (SCC) but not of BCC or melanoma.

Other researchers in the human trials of phase 3, double-blind, randomized, controlled trial, randomly assigned, in a 1:1 ratio of 386 participants who had had at least two nonmelanoma skin cancers in the previous 5 years to receive 500 mg of nicotinamide twice daily or placebo for 12 months witj participants evaluated by dermatologists at 3-month intervals for 18 months, suggested that treated patient showed a significant lowering nonmelanoma skin cancers by 23% at 12 months with compared placebo.

Both groups expressed  the similar differences with respect to new cases of basal-cell carcinomas and new squamous-cell carcinomas.
Also in compared to placebo, nicotinamide group showed a lower of number of actinic keratoses incidence, a scaly growth caused by damage from exposure to ultraviolet (UV) radiation  by 11% at 3 months (P=0.01), 14% lower at 6 months (P<0.001), 20% lower at 9 months (P<0.001), and 13% lower at 12 months (P=0.001).

Taking together, Niacin and it's dietaryNicotinamide may be a potential and therapeutic vitamin supplements for reduced risk and treatment for certain types of skin cancer.

All About Vitamins: Niacin and Schizophrenia

Niacin sensitivity and response may be associated to the developed schizophrenia, a renowned institute study suggested.
Niacin, is also known as vitamin B3, nicotinic acid, an organic compound with the formula
C6H5NO2, found abundantly in chicken, beef, fish, cereal, peanuts and legumes. It is best known for its effects in lowering cholesterol and triglycerides and removing toxic from our body and promoting production of steroid hormones.

Schizophrenia is a mental condition characterized by failure between thought, emotion, and behavior affecting patient ability in differentiated perception, appropriate actions and feelings......

 Schizophrenia patients showed a diminished neither sensitivity nor efficacy of niacin correlated with AA levels, in constrast to a significant correlation between AA levels and niacin sensitivity in control group according to the study of injection of eight concentrations of methylnicotinate to the forearms of 20 adults with schizophrenia and 20 controls,

The study also addressed that the levels of adrenic acid showed a strong correlated with niacin efficacy in schizophrenia patients and the correlation between adrenic acid and niacin efficacy in schizophrenia indicated other mechanism, such as expression of n-6 PUFA may involve to the blunted niacin response.

Dr. Messamore E, the lea author said, "The schizophrenia-associated niacin response abnormality involves both diminished sensitivity and reduced efficacy. The lack of expected correlation between levels of AA and adrenic acid suggests homeostatic imbalance within the n-6 polyunsaturated fatty acid (PUFA) pathway in schizophrenia".

Additionally, in the study of normal skin flush response, niacin attenuated in many individuals with schizophrenia (SCZ), showed an abnormal response to niacin, in inducing abnormal prostaglandin signaling due to depletion of AA, and other essential fatty acids (EFAs).

Other researchers insisted that the abnormal of niacin response of schizophrenia may also be associated to larger machenisms, including abnormalities in enzymes, receptors, or signal transduction mechanisms that affect the synthesis, release, or response to vasodilatory prostaglandins.

Taking together, there is no doubt that patient with schizophrenia is associated to niacin abnormal response and sensitive involved expression of many effects.

All About Vitamins: Vitamin B3 deficiency induced Dementia in Pellagra Patients

Vitamin B3 deficiency may be associated to the early onset and progressive risk of dementia in patients with pellagra, a respectable institute proposed.

 Dementia, the neurodegenerative diseases characterized by the loss of mental ability severe enough to interfere with people's every life. Alzheimer's disease is the most common type of dementia in aging people. About 5-8% of all people over the age of 65 have some form of dementia, and this number doubles every five years above that age.

Pellagra is a nutritional deficiency of tryptophan and niacin (nicotinic acid) disease

Niacin, is also known as vitamin B3, nicotinic acid, an organic compound with the formula
C6H5NO2, found abundantly in chicken, beef, fish, cereal, peanuts and legumes. It is best known for its effects in lowering cholesterol and triglycerides and removing toxic from our body and promoting production of steroid hormones.

According to the Kyoto University Graduate School of Medicine, dementia induced by patients with Pellagra, a photosensitivity syndrome postulated an exhibition of reactive oxygen species(ROS) production through increased levels of hormone prostaglandin E₂.

In niacin deficient mice model of pellagra, mice showed a significantly elevated inflammatory expression levels of COX-2, and PGE syntheses, in induced risk dementia.

In a case reported by the Anning Hospital, male with a classic case of pellagra bought to the hospital was found to process disorderly behavior induced cardinal symptoms of dermatitis, dementia caused by niacin deficiency.

The patients made a complete recovery after 6 months of appropriated treatment including supplement of niacin.

Additionally, in a case-control study of compared the past dietary habits of 342 Parkinson's disease (PD) patients recruited from nine German clinics with those of 342 controls from the same neighborhood or region, niacin intake through supplement or high niacincontent in coffee and alcoholic beverages, are found to have a inversely associated with PD.

The findings suggested that niacin plays a important role in reduced risks of pellagra expression of dementia and Pakistan disease.

All About Vitamins: Niacin and Risk of Depression

A 2015 study by a renowned institute suggested that niacin and other nutrient deficiency may be associated to a risk factor of psychological syndrome of depression.

Depression is a normal response as part of our daily lives such as the loss of s job, the death of a love one, and illness. Over 30 million Americans suffer from depression and the amount is increasing in an alarming rate.

Niacin, is also known as vitamin B3, nicotinic acid, an organic compound with the formula
C6H5NO2, found abundantly in chicken, beef, fish, cereal, peanuts and legumes. The vitamin is associated to cellular integrity and energy production involved in more than 500 intracellular reactions.

In a total of 130 older subjects participated in the study with 65 diagnosed depression (16 men and 49 women) and 65 age- and sex-matched controls without depression, the incidence of depression may be attributed the poor nutrients intake, including more intake of of sucrose and insufficient consumption of protein, fiber, eicosapentaenoic acid, niacin and vitamin B6.

The study also indicated that these nutrient inappropriate in patients with depression also expressed a lower quality of life in compared to control, by comprehensive geriatric assessment.

Other, in cross-sectional study conducted of the intake of major (that is, carbohydrates, fat, and protein) and minor (that is, vitamins and minerals) nutrients in adults with confirmed mood disorders, higher intake of nutrients are significant correlations to higher mental function and inversely associated to risk of mental disorder.

Mood disordered patients also showed a mild and moderated expression of depression, but not consistently.

Taking together, there is no doubt that deficiency of niacin other nutrients may exaggerated  the risk of depression.

All About Vitamins: Niacin in Prevention of Complication of Pellagra in Anorexia Nervosa

Niacin may be used as therapeutic form of treatment in patients of Anorexia Nervosa with complication of pellagra, a renowned institute insisted.

Anorexia nervosa is a type of eating disorder usually develop in the teen years and effect over 90% of female, because of excessive food restriction and irrational fear to become fat due the wrongly influenced media as attractiveness is equated to thinness.
Pellagra is a nutritional deficiency disease of tryptophan and niacin (nicotinic acid)

Niacin, is also known as vitamin B3, nicotinic acid, an organic compound with the formula
C6H5NO2, found abundantly in chicken, beef, fish, cereal, peanuts and legumes. It is best known for its effects in lowering cholesterol and triglycerides and removing toxic from our body and promoting production of steroid hormones.

According to the Canadian College of Naturopathic Medicine, the reviewed literature revealed several cases of pellagra in patients with anorexia nervosa whose cutaneous symptoms resolve, such as erythema on sun-exposed areas, glossitis, and stomatitis. were resolved within 24-48 hours after oral niacin administration of 150-500 mg.

Furthermore, there are other cases of a 14-year-old girl and 20-year-old woman with disorders of anorexia nervosa symptoms of pellagra were treated successfully with niacin therapy and dietary treatment.

Dr. Jagielska G, the lead author in the study conducted by the Medical University of Warszaw, said, " Pellagra is a potentially fatal, nutritional disease with cutaneous, gastrointestinal, and neuropsychiatric manifestations. Because of the diversity of pellagra's signs and symptoms, diagnosis is difficult without an appropriate index of suspicion".

The finding evidences suggested restriction diets in patients diagnosed with anorexia nervosa may cause significant nutritional deficiency, inducing Pellagra, although it is not common.
